Single-nanopore based channel current analysis and detection. A nanometer-scale channel can be used to associate ionic current measurements with single-molecule channel blockades (Fig. 1, Top). The α-hemolysin channel self-assembles, leading to an inexpensive and reproducible nanopore detector. The α-hemolysin based nanopore detector is well-suited to observation of biomolecular kinetics that can modulate ionic flow through its limiting aperture.
