Affinity purification experiments using P83916 (CBX1), Q99496 (RNF2), P16104 (H2AFX), and Q09028 (RBBP4) as baits. (A) Immunoblot confirming the expression of the indicated FLAG-tagged chromatin proteins using antibody against the 3X FLAG epitope. Two independent FLAG tag constructs of each chromatin related protein was constructed for affinity purifications to eliminate background contaminants and to uncover highly reproducible interactions. Molecular masses (kDa) of marker proteins by SDS-PAGE are indicated. (B) Representative functional clusters identified from affinity purification data (light gray) and expanded by MP-PIPE predictions (dark gray). Tagged-baits are shown by yellow nodes (ellipses).
