Substraction of background fluorescence. Upper: Calibration curve constructed from the fluorescence and absorbance data in well A3, defined as the background for wells A5-A12. The calibration curve is a smoothing spline with a user-defined smoothing parameter. The curve can be extrapolated outside the range of observed absorbance values, a feature that is not needed in this experiment.
